桃红颈天牛雌雄成虫转录组差异分析

中文摘要:为明确桃红颈天牛Aromia bungii雌雄成虫转录组中的差异表达基因及其相关功能,采用高通量测序技术对雌雄成虫进行转录组测序,并对测序结果进行功能注释分析和差异表达基因的GO功能和KEGG通路富集分析。结果显示:从桃红颈天牛雌雄成虫转录组数据中共获得31 077个unigene,有21 613个unigene在NR、KOG、KEGG、GO、SwissProt、NT和PFAM七大数据库得到注释,其中在NR数据库获得注释的unigene最多,达18 201个。桃红颈天牛雌雄成虫间存在6 944个差异表达基因,其中雌成虫中相对于雄成虫显著上调基因有3 309个,显著下调基因有3 635个。基于差异表达基因的GO功能和KEGG通路富集分析,挖掘出8个可作为新型防治靶标的生殖相关基因,分别为卵黄原蛋白基因、卵黄原蛋白受体基因、类固醇急性调节蛋白基因、细胞周期蛋白B3基因、精液蛋白基因、精子发生相关蛋白6基因、透明质酸酶基因和cAMP依赖性蛋白激酶基因。

Transcriptomic analysis of differential gene expression between female and male adults of red-necked longicorn Aromia bungii

Abstract:To identify differentially expressed genes (DEGs) and their associated biological functions between male and female adults of red-necked longicorn Aromia bungii, high-throughput transcriptome sequencing was conducted. The sequencing data were subjected to functional annotation and enrichment analyses based on Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) databases. A total of 31 077 unigenes were assembled, of which 21 613 were successfully annotated across seven major databases (NR, KOG, KEGG, GO, SwissProt, NT, and PFAM), with the NR database yielding the most annotations (18 201 unigenes). Comparative analysis revealed 6 944 DEGs between male and female adults, including 3 309 genes significantly up-regulated and 3 635 genes were significantly down-regulated in females relative to males. Functional enrichment analyses of these DEGs led to the identification of eight reproduction-related genes with potential as novel targets for pest control, including genes encoding vitellogenin, vitellogenin receptor, steroidogenic acute regulatory protein, cyclin-b3, seminal fluid protein, spermatogenesis-associated protein 6, hyaluronidase and cAMPdependent protein kinase.
